We always travel with fresh water full, grey/black empty on way out. I also try to keep gas tank and propane full.
Coming home, grey and black are usually 1/2 to full.
Weight is no issue, the e450 list cargo capacity of 3199 lbs. We have been able to get unit to 13,400 plus 5200 lb trailer. even carrying extra 50 gallons of fresh water in RV, tools and parts. We never get to the 14,500 GVWR/22,000 GCVW limits.
We did a lot of math homework prior to purchase to be certain towing/loading would no be issue.
Performance of V-10/6 speed has been impressive. I know the diesels pull harder but I never have issues merging passing with or without trailer.
