I purchased the valve on the top to replace the regulator on the bottom, the bottom reg/control is the original to my grill. I also purchased a 12' hose at the same time.

propane hookup

I have a hose that connects to rv quick connect & my Coleman grill without regulator. Works great. Now I want to hookup 20lb tank to rv for when parked for a long time. Will it work with a hose with regulator hooked up to rv quick connect whose hose goes to line that goes inside rv & is connected AFTER the rv's regulater? With the rv;s tank turned off.

No - using portable tanks to supplement the onboard tank is done on the high pressure, non-regulated side. Extend-a-stay is one brand adapter for this purpose.
