Our Colorado requires disconnecting the battery due to the electric assist power steering which must be disabled while towing. But the brake system does require 12 VDC to operate so I installed a remote disconnect on the positive side of the battery which also provides a separate power wire from the battery for use by the breaking system. No charging wire required.

I don't know what the difference is on a Gladiator. I'd follow the owner's manual.
I just completed the brake and tail light wiring, Demco Stay-In-Play Duo install in my Jeep JL. And I ran a charge wire W/Diode.
My owners manual states for towing behind a MH, transfer case in neutral, auto trans in park and away we go.

Same exact installation for the JT. It's basically a stretched Wrangler with a 5' pickup bed. And same setup; transfer case to N, tranny to Park, flip switch on the Demco to ON, and we're off and running. No disconnecting the battery.

I have a 2021 Gladiator and nowhere does it say to disconnect the battery. A friend with a 2020 gladiator, her manual says disconnect the battery, but it doesn't say disconnect the second battery for the autostart, which caused them some problems, I believe it has to do with the wiring harness that is used and where you get the power fir auxiliary brake.
