Jamie, there is very little difference in top overdrive gear ratio between the 5- and 6-speed transmissions, although the new 6R140 is reportedly slightly more efficient for other reasons. However, I can't see anything that would explain 20% or greater MPG improvement.
Final gear ratio could make a bigger difference since in theory it could vary from 4:10 to 4:56, but even that is unlikely to result in such large MPG improvement. I expect all newer E-450s have the 4:56 gears anyway.
A compact Class A that could deliver 12 MPG consistently on the highway would be pretty nice.
